Biofilme Estafilocócico: uma fonte de prejuízos para a pecuária leiteira na região agreste de Pernambuco, Brasil

Abstract

No Brasil, a pecuária leiteira sofre uma perda anual equivalente a 2 bilhões de litros de leite (5% da produção anual), por conta da mastite bovina. Isso equivale a um prejuízo de R$ 1000 (US$ 184) por vaca ao ano. A bactéria Staphylococcus aureus é um dos principais agentes causadores da mastite, infecção da glândula mamária. A cronicidade da mastite está ligada a diversos fatores de virulência presente em S. aureus, dentre eles destaca-se a formação de biofilme, associada à presença dos genes do cluster gênico ica, facilitando sua adesão e permanência no tecido mamário, além de proteção contra a ação do sistema imune do hospedeiro e de antimicrobianos. O objetivo deste trabalho foi analisar a produção de biofilme por S. aureus isolados de leite cru e queijo coalho produzidos e comercializados no Agreste Meridional de Pernambuco. A partir das 53 amostras analisadas, foram isoladas 19 cepas de S. aureus, que foram caracterizadas quanto a produção de biofilme in vitro, utilizando dois meios de cultura diferentes (BHI e TSB + sacarose 1%). Estes isolados também foram investigados quanto a presença dos genes icaA e icaD por PCR. Das 19 cepas, 63,2% (12/19) apresentaram produção de biofilme, variando entre forte (6/12) moderada (2/12) e fraca (4/12), enquanto 36,8% (7/19) não produziram biofilme. Na análise molecular, os genes icaA e icaD foram detectados em 100% dos isolados. A expressão desses genes têm impacto significativo no desenvolvimento e manutenção da mastite estafilocócica bovina, sendo necessária medidas mais efetivas de monitoramento, controle e profilaxia desta afecção.
